Biomolecular Discovery Services now offers next-generation sequencing (NGS) in combination with selections based on synthetic, naïve or custom-immunized VHH phage display libraries. Instead of tens or even hundreds of possible leads, NGS provides you with tens of thousands of sequences rank-ordered by their prevalence in the selection. This is especially important when there are immunodominant sequences that repeatedly arise from standard selections.
By making parallel selections, you can discover VHH in common epitopes and unique epitopes and eliminate VHH in tags or fusion proteins. Combined with affinity maturation selections, you can find which residues are critical for improved binding. By altering binding conditions, you can determine which residues the change affects (salt, temperature, epitope mutations).
